Development of an efficient protocol for the production of deer embryos by in vitro fertilization: Application of sexed semen

The general objective of this project is to develop a suitable protocol for in vitro fertilization in deer and adapt to the use of sexed sperm samples in this species. For this we propose to develop the following specific objectives:

1. To study the ideal time to capacitate spermatozoa and develop defined medium for the capacitation;

2. To determine the effect of oxygen tension during maturation and in vitro fertilization of oocytes on embryonic development;

3. To assess the effect of the absence or presence and the optimal time of addition of fetal bovine serum for in vitro culture of embryos on embryonic development;

4. To study conditions and optimal capacitation time for sexed sperm and the optimal concentration of sexed sperm in IVF.
